SUMMARY:Clinical Leader - Cancer Precision Medicine
DESCRIPTION:The UNC Lineberger Comprehensive Cancer Center and The University of North Carolina School of Medicine seek an exceptional physician scientist to lead our efforts in Cancer Precision Medicine. Over the past decade\, UNC Lineberger emerged as a leader in the NCI TCGA effort. We have developed an infrastructure for sequencing\, bioinformatic analyses\, molecular pathology\, and their application to patient care through a molecular tumor board. We now search for a translational scientist to head the clinical aspects of an expanding program. We are currently developing novel CLIA assays as well as methodologies to perform genomic analysis to identify somatic and germline mutations\, evaluate transcriptome expression using RNA-sequencing\, and characterize immune repertoires using next generation sequencing. We have developed robust applications for data analysis and anticipate that the person leading this group will utilize these other tools they develop to enhance personalized care for cancer patients and prospective clinical trials. The position is tenure track and open to those at all ranks. Applicants should provide evidence of a strong record of clinical\, translational or basic approaches to broad areas of genetics\, genomics\, gene expression\, epigenetics\, proteomics\, tumor immunology\, and/or precision early cancer detection and prevention. SUMMARY:Research Scientist - biology & developmental therapeutics
DESCRIPTION:Dr. Kapil N. Bhalla is currently seeking a Research Scientist who will be responsible for conducting research in his NIH-funded laboratory. The focus of the basic and translational research in this laboratory includes the biology and developmental therapeutics related to epigenetic mechanisms involving histone deacetylases\, methyltransferases and demethylases\, BET proteins\, as well as the chaperone biology of heat shock proteins and their cancer relevant client proteins regulating cell growth\, differentiation and survival of the human leukemia stem/progenitor cells. \n\nThe preferred candidate will possess a Ph.D. in Biology\, Biological Sciences\, Life Sciences or related field; a keen interest and experience in cancer research and excellent English skills (both written and verbal).  The preferred candidate will have completed at least a 3 year Post-doctoral fellowship in an area relevant to the focus of research in Dr. Bhalla’s laboratory. In addition\, have the ability to troubleshoot in the design and conduct the laboratory studies. Experience in molecular studies involving gene transfection\, analysis of gene expression through QPCR\, microarrays\, immunoblots and flow cytometry\, as well as experience in chromatin immunoprecipitation studies\, evaluation of protein biochemistry with 2D gels and the yeast-two-hybrid system\, and tissue culture techniques is required. Experience in the ability to utilize CRISPR technology for gene editing and to utilize confocal microscopy for molecular imaging is highly desired. Experience in xenograft and genetically-engineered animal models of cancer and/or leukemia\, and confocal microscopy is also desired.  An outstanding research environment\, new laboratory and core facilities are provided. \n\nKEY FUNCTIONS \n  \n1. The basic and translational research focus of this laboratory includes epigenetic mechanisms involving histone deacetylases\, methyltransferases and demethylases\, heat shock proteins and their cancer and leukemia-relevant client proteins\, as well as transfection and functional analyses of genes regulating cell death/survival of human cancer cells. Experience in animal studies involving NOD/SCID or nude mice\, and mouse imaging (xenogen and MRI) is highly desirable. Experience in molecular techniques involving plasmid DNA preparations and transfection\, the analysis of gene expression including QPCR\, microarrays\, chromatin immunoprecipitation\, evaluation of protein biochemistry with 2D gels\, animal models\, the yeast-two-hybrid system\, confocal microscopy and tissue culture are also preferred. \n2. Plans\, organizes\, coordinates\, directs\, and participates in scientific research projects.  \nPerforms quality research in relation to current research projects\, and other identified needs. Interprets results of experiments\, and makes necessary changes in procedures when needed.  Compiles data sheets. \n3. Organizes and performs routine procedures involving molecular biology and biochemistry of RNA processing. Requires experience in tissue culture\, DNA & RNA preparation\, PCR\, DNA cloning\, protein synthesis and electrophoresis. \n  \nResponsible for providing laboratory services\, tissue culture\, processing research samples\, and/or cataloging samples as required. Interacts with the principal investigators\, postdoctoral fellows and other research personnel in communicating experimental design and results. \n4. Requires knowledge of standard radiographic techniques. Dark room usage is essential as is use and proper handling of radioisotopes. \nMust be able to comply with all institutional safety policies including Universal Precautions and OSHA standards when handling toxic\, hazardous\, flammable\, or potentially infectious or carcinogenic materials.  This may require wearing common protective garments or equipment such as gloves\, eye protection\, face mask\, gown or lab coat\, etc.  Additionally some procedures may require the use of a fume hood or biological safety cabinet. \n5. Work with laboratory animals such as mice and rats and to perform interventional procedures such as tail vein injections and orthotopic implantations. Retrieves\, maintains\, and catalogs specimen stocks from outside reference laboratories.  Assists in other laboratory activities as needed. \n6. Performs and coordinates scientific experiments in a collaborative effort with other technicians\, postdoctoral fellows\, and staff members.  Monitors work flow as it pertains to the need for supplies\, equipment maintenance and stock solutions.  Performs preventive maintenance on laboratory equipment. \n7. Equipment monitoring on a daily basis verifying correct temperatures of freezers\, refrigerators\, incubators\, etc.  Equipment maintenance on a weekly basis includes cleaning and disinfecting refrigerators and incubators.  Daily counter clean up and disinfection to ensure safety of the laboratory work place.  Bags waste and transports to appropriate refuse area utilizing safety standards and guidelines.  Maintains all safety precautions. \n8. Assigns and reviews work of subordinates\, determines specific goals or objectives to be attained\, evaluates resulting research data\, and revises techniques or approaches to work problems. Supervises the design\, development\, or adapting of equipment used in experiments or research to obtain desired results. Utilizes institutional and departmental databases to perform data retrieval for analysis.  Designs retrieval templates for standard queries.  Apply bio-statistical methods in analyzing clinical data such as pretreatment characteristics\, response to treatment\, toxicities\, and mortality\, etc.  \nConducts Library Work.  Finds and copies journal articles as required.  Reads and understands articles related to ongoing research.  Maintains references in a manual filing system.  Requires good reading and comprehension skills as well as diligence\, patience and ability to carry several heavy books at a time. \n9. Assemble samples\, reagents\, dry or wet ice\, data and packaging materials; draft a cover letter\, accurately and legibly fill out shipping labels; carry package to receiving area on ground floor prior to pick up.  Keep accurate legible logs of outgoing and incoming packages.  For incoming packages\, ascertain contents\, store at appropriate location\, file information sheets and notify principal investigator.  Update pertinent inventory logs. \n10. Compiles\, writes\, and submits reports to superior.  Plans and supervises the training of technicians and other laboratory personnel with respect to proper laboratory techniques\, the use of laboratory equipment\, and safety procedures. \n11. Presents findings of data to PI and prepares scientific documents for publication.  Presents findings of research projects to professional groups and prepares scientific documents for publication.  \n12. The Gardner lab studies basic mechanisms of epigenetic gene regulation in breast cancer. The lab approaches basic questions in breast cancer biology with an integrated interdisciplinary focus on fundamental mechanisms of chromatin-based transcriptional control\, epigenetic regulation and the influence of the environment diet and metabolic imbalance (Proc Natl Acad Sci U S A. 2009 Nov 17;106(46):19286-91; Nat Struct Mol Biol. 2010 Dec;17(12):1406-13; Nat Commun. 2012 Jan 17;3:633; Nat Commun. 2013;4:1449). Major goals include understanding the role of metabolism and the transcriptional cross-talk between epigenetic co-regulators and hormone receptor-mediated signaling pathways in both enhancer function and promoter-targeted mechanisms of gene control. An essential component of this work will involve developing omic perspectives that will integrate next-generation sequencing with genomics\, transcriptomics\, epigenomics\, proteomics and metabolomics using model systems and patient-derived samples. The projects will be supported by access to an established cohort of diverse breast cancer patient samples with extensive molecular and genomic characterization to aid in developing translation perspectives in gene-network and pathway discovery that will have practical implications for improving breast cancer diagnosis\, treatment\, and prevention. \nPostdoctoral experience and an extensive background in bioinformatics including high throughput analysis pipelines such as\, ChIPSeq\, RNASeq\, ExomeSeq\, microarray analysis\, peak calling and comparative enrichment analyses. The candidate will provide support for routine CHIPSeq\, DNAse Hypersensitivity\, ATAC-seq and MethylIP Analysis and provide bioinformatics support on DNA motif analysis. The candidate should have expertise in both peak calling of ChIP-seq data as well as RNA-seq analysis for splicing changes. Experience with off the shelf tools\, including Bowtie\, Scripture\, Cufflinks\, TopHat\, SICER\, and MI Knowledge in statistics is essential. Proficiency in one\, preferably two\, of the following programming languages: Shell\, Perl\, Python\, R\, Java and C/C++ is essential. Experience and familiarity with public databases: NCBI\, Ensembl\, TCGA\, cBioPortal\, Broad FireHose is essential. Central goals in the projects are to identify meaningful genome-wide or locus/DNA-feature-specific changes. The ability to generate novel scripts for specific applications\, including SNP calling for allele-specific effects and distance mapping for positional effects is highly desired. Experience in applied statistics and design of experiments is desirable. Experience in submitting data sets to public repositories is required. DESCRIPTION:The research to be carried out focuses on the micro-environmental and epigenetic regulation of disseminated cancer cell dormancy and metastasis initiation. The project will focus on the mechanisms by which signals from the micro-environment reprogram tumor cells into dormancy. The project will focus on identifying signals produced by organ specific macrophages that impinge on solitary tumor cell behavior and how it impacts the epigenome of the cancer cells. \nThe candidate should have experience in basic molecular and cellular biology methods including DNA and RNA isolation\, siRNA methods\, construct engineering\, tissue culture and preparation and analysis of samples through flow cytometry. The experiments will also require biochemistry techniques including Western blots and immuno-precipitations among others. These will focus primarily on ChIP-seq and ATAC-Seq when necessary. Expertise in code writing and/or bioinformatics will be weighed as a plus. Single cell RNA-seq will be part of the research efforts as well. The in vivo aspects of the work will entail growing human cancers and detecting single solitary DTCs using microscopy or single cell isolation tools\, metastasis in specific pathogen-free embryonated egg chorioalllantoic membrane system and performing experiments with transgenic mice and nude mice. Effort will also be allocated to immuno-histochemistry and immuno-fluorescence techniques. \nThe candidate will report directly to the PI and will collaborate with other postdoctoral fellows and students on other projects as directed by the PI. The Aguirre-Ghiso lab engages in trans-Atlantic\, regional and national collaborations and thus the candidate will have to be amenable to traveling when needed and interacting in a collaboratively and proactive manner with collaborators of the Aguirre-Ghiso Lab. It is anticipated that the candidate will be open to learning new techniques and methods that will fulfill the varying needs of research projects in the lab. It is anticipated that the postdoctoral fellow to be hired will maintain fluid and straightforward communication with the PI. It is expected that the candidate will maintain normal working hours with a presence in the lab during the majority of the working hours of the day and that any change in schedule will have to be approved by the PI and submitted with time in advance for approval.

SUMMARY:Postdoctoral Fellow: ­­­­­­­­­­Cancer Immunotherapy
DESCRIPTION:Cancer immunotherapy represents a paradigm shift in the management of cancer. Recent successes with antibody directed therapies for immune modulators such as CTLA4 and PD1/PDL-1 axis have altered the landscape of systemic therapy for solid and hematologic malignancies. Among various strategies to improve cancer immunotherapy\, epigenetic targets have emerged as prominent actionable targets to enhance anti-tumor immune responses and suppressing tolerogenic tumor microenvironment. Our laboratory is focused on mechanisms by which various epigenetic molecules impact the immune system to identify novel approaches to cancer immunotherapy. \n  \nAlternative approaches to enhancing cancer immunotherapy are to modulate immunologic consequences to existing cytotoxic therapy\, such as radiation therapy. More recently\, the concept of immunogenic cell death induced by XRT has been proposed. Immunogenic cell death is associated with release of HMG-B1 and ATP as DAMPs (damage-associated molecular pattern)\, which provides the “Danger Signal” required for effective adaptive immune responses. Surface translocation of calreticulin promotes antigen presentation by facilitating phagocytic uptake by dendritic cells (DC). We are therefore currently investigating how to promote tumor specific immune responses by radiation therapy. \n  \nPosition Highlights: \n\nInvolved mentorship by Dr. Kim\n\n\nGain knowledge base in cancer immunotherapy\, immune activation and tolerance mechanisms\, molecular immunology\nDevelop expertise in epigenetic regulation of the immune system\nTrain in in vivo and in vitro methodologies in immunologic research\n\n  \nThe Ideal Candidate: \n\nScientifically inquisitive and enthusiastic to learn new approaches\nCollaborate well with other researchers\nBackgrounds in immunologic research\nFamiliar with flow cytometry and molecular work\n\n  \nResponsibilities: \n\nRead and discuss relevant literature\nDesign and conduct experiments\nPresent at the lab meetings and scientific conferences\nResearch methodologies will mostly include flow cytometry\, confocal microscopy\, proximity ligation assay\, Western blot and other molecular work\, ex vivo and in vivo mouse studies.\n\n  \nCredentials and Qualifications: \n\nPh.D. or M.D. in Immunology\, Molecular Biology\, Cell Biology\, or Cancer Biology.\n\n  \nA competitive salary and benefits package will be offered to the successful candidate.
