DESCRIPTION:NIH funded postdoctoral position available immediately at Northwestern University in the Feinberg School of Medicine in the Department of Urology. The postdoctoral fellow will work closely with Joshua Meeks\, MD\, PhD in the Department of Urology and Stephen Miller\, PhD in the department of Microbiology and Immunology. Research projects associated with this fellowship include: 1) mechanisms of T-cell biology and immune activation in genitourinary disorders (including cancer)\, 2) mechanisms resistance to therapy\, 3) mechanisms of T cell tolerance 4) epigenetic regulation of T cell activity 5) novel checkpoint and immune regulatory therapies. \nCandidates are required to have US citizenship or permanent residence and a PhD or MD in Molecular biology\, or Immunology and/or comparable experience in these subjects. DESCRIPTION:ECLIPSE (Evolution of Cancer\, Leukemia and Immunity Project) is a novel organization within the MD Anderson Cancer Center\, Houston\, Texas\, with the mission of making MDACC and its partners recognized leaders in the identification and development of innovative immunooncology therapies that will cure all hematologic malignancies. \nThe primary requirement for an individual in this position is an experienced and highly motivated individual with hands-on experience in identification and validation of potential therapeutic targets for patients with advanced hematological malignancies. Such data generated from the team is expected to influence a broad array of decisions around target prioritization and generation of therapeutic modalities against those particular targets. The successful candidate will participate in cross-functional research to design and develop novel assays that will support an understanding of target biology. The selected candidate will be expected to work at the bench and contribute to development and implementation of potential biomarker strategies to support clinical development. Previous experience in biotech/pharma is highly preferred. \n\nKey Functions \n· Design and develop key experiments contributing to the conception\, execution\, and championing of novel targets\, technology platforms\, and strategies related to the development of biologics-based clinical candidate programs. \n· Provide expert technical guidance to others on project/task groups. Within each these areas\, discovery research\, translational research\, and early clinical development are integrated into a single team.  \nSummary/Scope:\nThe PH TCoE is built on our legacy IMiD drugs\, which exhibited therapeutic efficacy in erythema nodosum leprosum\, myelodysplastic syndrome\, and multiple myeloma. This class of drugs hijack the cereblon E3 ubiquitin ligase to eliminate disease-driving proteins that are otherwise considered “undruggable”. The early drug discovery team in the PH TCoE is now seeking a motivated PhD scientist to assist the development of cereblon modulating agents that target novel cereblon neosubstrates for ubiquitination and degradation for the treatment of a variety of human diseases. The successful candidate will develop and utilize state-of-art biotechnology tools to identify novel substrates and perform target validation for the relevant disease indication. DESCRIPTION:A postdoctoral position is available in the MacPherson Lab at Fred Hutchinson Cancer Research Center. The project will focus on small cell lung cancer (SCLC)\, a neuroendocrine tumor type that exhibits initial sensitivity to chemotherapy but invariable emergence of chemoresistance. Our lab is interested in linking specific gene mutations in SCLC to therapeutic vulnerabilities and to chemoresistance. We have developed a panel of new genetically engineered mouse models (e.g. see Jia et al\, 2018 Cancer Discovery) and patient derived xenograft models that can be used in these efforts. Many projects in the lab incorporate genome-wide functional screens in cells and in vivo. These screens are used to identify functionally important genes for SCLC and genetic vulnerabilities that might lead to new avenues to therapeutically target SCLC. One available project includes interrogating the importance of candidate SCLC driver genes and linking specific SCLC-mutated genes to novel therapeutic approaches. Hits identified through functional screens are followed by therapeutic studies that employ GEM and PDX model. A second available project applies in vivo genome-wide screens to understand drivers of chemoresistance.\n\n\n\nQualifications\n\n\n\nWe are looking for a highly motivated individual with a PhD or MD and interests in cancer genetics\, epigenetics\, mouse models\, functional screening and/or therapeutics. DESCRIPTION:The Souroullas Lab studies the role of chromatin dynamics during cancer development\, with a specific focus on hematopoietic (lymphoid) malignancies and melanoma. Recent next-generation sequencing studies have highlighted the role that epigenetic modifiers play in cancer\, however\, our understanding of these mechanisms remains quite rudimentary. We have recently demonstrated the oncogenic and neomorphic activity of mutations in the EZH2 gene\, part of a protein complex which mediates histone methylation and regulates gene expression (Souroullas et. al.\, Nature Medicine\, 2016). The underlying mechanisms of the oncogenic activity of these mutations\, however\, remain unresolved. Research opportunities are therefore available for highly motivated individuals with an interest in cancer genetics and/or epigenetics. Projects include\, but are not limited to\, the study of genetic interactions using mouse models\, molecular analysis of epigenetic crosstalk\, and the effects of these mutations on the tumor immune response. DESCRIPTION:The successful candidate will join the Epigenetic Etiology of Human Disease Laboratory to study the role of DNA epigenetic modifications in human disease. We are using a variety of in vivo and in vitro model systems to study the functional consequences of RNA splicing factor and epigenetic modifier mutations on stem cell function and to identify new targeted therapies for myeloid diseases.
