DESCRIPTION:A Postdoctoral Associate position is available in the laboratory of Alexey Terskikh\, Development\, Aging\, and Regeneration Program. The position focuses on the novel approach to study the dynamics of 3D epigenetics landscape at a single cell level. The Postdoctoral Associate will explore the capability of Microscopic Imaging of Epigenetic Landscape (MIEL) and fluorescence-based reporters enabling live imaging of epigenetic modifications at a single cell level. MIEL combines fluorescence detection of epigenetic marks with automated cell imaging and machine learning to profile and quantify the “epigenetic signatures” of individual cells. \nThe candidate will investigate the epigenetic heterogeneity at the single cell level and the asymmetry during stem cell division in normal neurogenesis and in brain tumors. One of the long-standing question of this project is to determine whether the outcome of stem cell division is stochastic or deterministic and whether such outcome could be predicted based on the epigenetic signature of the mother cell. \nEducation and/or Experience: \nInterested candidates must have a Ph.D. degree in any discipline of Biological Sciences. SUMMARY:Post Doctoral Scientist
DESCRIPTION:A Postdoctoral Scientist position is available for a skilled and motivated scientist in the Laboratory of Dr. Maho Shibata in the Department of Anatomy and Cell Biology and the Cancer Center at the George Washington University School of Medicine and Health Sciences. \nDr. Shibata’s lab applies concepts from developmental biology and stem cell biology to prostate cancer research. The lab uses genetically engineered mouse models\, explant cultures\, and 3D organoid culture systems to study androgen signaling during prostate development\, and to identify novel molecular drivers of castration-resistant prostate cancer. There are multiple project possibilities. \nDuties include: \n\nIndependently design experiments\, collect and analyze data\, and assist collaborators\nPresent findings in group meetings and internal and external scientific meetings\nPrepare and submit postdoctoral fellowship applications\nWrite and publish research manuscripts\nSupervise and train students\nPerform other duties as assigned. The omission of specific duties does not preclude the supervisor from assigning duties that are logically related to the position.\n\nOur lab is located in the George Washington University (GW) Cancer Center in the highly urban center of Washington DC\, on the top floor of the state-of-the-art Science and Engineering Hall\, which opened in 2016. The GW Cancer Center brings together cancer research\, clinical cancer care\, and cancer control/prevention and outreach initiatives at GW. The District of Columbia faces prominent health disparities in several cancers\, including prostate cancer. For example\, cancer mortality among African Americans in D.C. is 10.9% higher than the national average. D.C. also has the highest percentage of individuals identifying as lesbian\, gay\, bisexual or transgender (LGBT\, 10% of the District’s population) when compared to the 50 U.S. states\, and the largest number of same-sex couple households. GW is developing initiatives that address cancer within these communities\, and will endeavor to become a national leader in advocating for policies to better understand and address health disparities.\nGW has demonstrated a strong commitment to cancer research by recruiting expert researchers in establishing the GW Cancer Center to grow and diversify cancer research locally. DESCRIPTION:Application deadline: January 15\, 2019 \nReports to the Director of the Cancer Center and the Executive Director of the Cancer Center\, and is responsible for administration of all research programs within the MGH Cancer Center. Works closely with the Cancer Center’s Scientific Director\, Clinical Director and Director of Clinical Research on the oversight and integration of laboratory-based\, translational and clinical research. \nDue to rapid advances in scientific discovery and clinical trials at the MGH Cancer Center\, there is an increasing need to provide strong administrative and strategic direction and to develop formal linkages between a large array of basic\, translational and clinical research programs on cancer. The MGH is committed to speeding the development of new diagnostic tests\, cancer treatments\, and other interventions that benefit individuals with cancer and people at risk for developing cancer. Both existing and planned cancer research efforts will require formalized administrative support and oversight to achieve our goals of scientific discovery and their clinical applications. \nKey to multi-year initiatives will be the development of an organizational structure to support current and future needs of the MGH Cancer Center research programs. New research ventures include collaborations related to genomic\, epigenetic\, proteomic\, chemical biology\, immunobiology\, and computational biology resources; and multi-disciplinary programs in liquid biopsies and early cancer detection\, cancer immunotherapy and immunopathology assays\, and both inter-academic consortia and pharma/biotech alliances. \nIn addition to major responsibilities for administration of the research mission within the MGH Cancer Center\, the position also serves as an institutional representative to the Harvard-wide comprehensive cancer center (seven-institution consortium Dana Farber/Harvard Cancer Center; DF/HCC)\, representing the MGH Cancer Center for all basic and translational research activities\, including Harvard-wide clinical trials. In addition\, major translational efforts link the basic and clinical enterprises\, including the CAR-T/immune cell processing facilities and tissue-based collaborations with the MGH Pathology Department\, and the Cancer Center serves to coordinate and support cancer research housed in multiple individual MGH Departments. Key roles and responsibilities include the following: \n\nProvide oversight of finances\, space\, operations\, and development for all Cancer Center research programs\, in concert with the Cancer Center leadership and the associated Principal Investigators.\n\n\nResponsible for strategic planning for future collaborations between academic partners and industry.\n\n\nIn concert with Cancer Center leadership\, develop the strategic plan and business case for basic\, translational and clinical research initiatives and work with the Cancer Center Director and Executive Director to align institutional resources and fundraising priorities.\n\n\nWork closely with the Cancer Center and Clinical Director on recruitments of research faculty and staff including the development of support packages.\n\n\nDevelop implementation plans for new collaborations and interfaces with Legal Counsel\, the Office of Industry Interactions\, and Hospital administration.\n\n\nInterface with the IRB and monitor potential conflicts of interest; also work with the Cancer Center Practice Administration to operationalize research efforts within the clinical setting; oversight over lab-based operations and research grant management.\n\n\nWork with Program Directors to balance priorities\, clinical trial portfolios\, and staffing.\n\n\nDevelop strategies for clinical trials in the network and support clinical trials advisory services as part of international advisory engagements.\n\nThe Director of Research will also serve as an institutional liaison to DF/HCC consortium and work with the Cancer Center Director on matters related to governance\, participation\, and fulfillment of grant requirements. The Director of Research will have direct oversight over the scientific Senior Project Managers and the research grant management team supporting the Center for Cancer Research\, which constitutes the lab-based faculty. He/she will also have oversight over the Administrative Director overseeing the Cancer Center Protocol Office (CCPO)\, which manages all cancer-related clinical trials at MGH\, including data management\, clinical research support and grants administration. The Cancer Center Grants Management team report directly to the Director of Research; and both Development and Marketing/Public Affairs functions associated with cancer research will have a matrix report to the Director of Research. \nLaboratory Research: The CCR is an inter-deparmental research unit that includes approximately 50 independent faculty members\, with academic appointments in the various departments that constitute the Cancer Center\, some 300 laboratory members (postdoctoral fellows\, graduate students and technologists) approximately 60\,000 sq ft of research space and an annual research budget of ~$50M. Administrative staff include a Scientific Programs Manager\, multiple lab managers\, research grant administrators and administrative support staff. The Director of Research will oversee the administrative components of this large\, interactive and vibrant research community\, working directly with the Cancer Center Director and Scientific Director to ensure continued academic success and financial strength. In addition to individual laboratory groups\, the CCR includes core resources\, and supports translational facilities\, many of which are in partnership with the Dept of Pathology\, requiring both fiscal and administrative oversight. \nClinical Research (Protocol Office): Much of the clinical research program for the MGH Cancer Center is centered around the Cancer Center Protocol Office (CCPPO) which coordinates the development of both interventional and non-interventional research studies in cancer. The CCPO employs approximately 150 data managers and research nurses\, with an annual budget of ~$15M\, derived in part from pharmaceutical contracts\, research grants\, philanthropic donations\, and institutional support. The Administrative Director of the CCPO\, who oversees all operational aspects of the CCPO\, will report directly to the Director of Research. In addition\, the Director of Research will work with the Clinical Director and the Director of Clinical Research to formulate and implement the overall strategy for the MGH Cancer Center research portfolio\, pharma/biotech research alliances and partnerships. This will also include working with the CCPO Administrative Director to implement management strategies\, quality assurance methods and develop tools for financial analysis. Together with the CCPO Adminsitrative Director\, the Director of Research will also provide input on the feasibility of conducting trials\, potential overlap or conflict with ongoing trials\, and logistical and administrative issues; maintaining protocol information management and analysis system; acting as liaison with national cooperative groups\, and maintaining information on investigational drugs\, devices\, therapies and diagnostics used by investigators\, consistent with audit requirements. \nClinical Research (DF/HCC CCSG): While the implementation of clinical cancer research at MGH is managed through the MGH CCPO\, the MGH is a founding member of the DF/HCC comprehensive cancer center grant (CCSG) consortium\, and the mandated IRB (institutional review board) and SRC (scientific review) are jointly administered by the three clinical institutions (MGH\, Dana Farber/Brigham\, and Beth Israel Deaconess) through the DFCI IRB. The Director of Research will serve as a member of the DFCI Institutional Review Board\, and have the key role of representing the MGH clinical research enterprise in the joint CCSG. In addition\, the Director of Research will participate in the MGH Clinical Research Operations Committee\, the MGH Clinical Policy and Oversight Committee (ClinPOC) and the DF/PCC Network Affiliate Committee. He/she will assists with the development of standards and procedures to coordinate clinical research effort across MGH and its partner institutions\, including strategic planning of clinical research programs at MGH affiliated sites\, foreign and domestic. The Director of Research will also have administrative responsibilities over the Cancer Data Registry\, which encompasses the Cancer Data Registries at MGH and MEEI\, working with the Manager of the Cancer Data Registry to devise and implement management and quality assurance strategies; providing budgetary oversight for the Registry\, including capital and personnel resources to support office operations.; working with the Medical Director to ensure that standards of the State Department of Public Health are met. \nAdministration and Supervision: The Director of Research will have management oversight of the Administrative Director for research grants and finances\, the Administrative Director of the CCPO\, the Manager of the Cancer Registry\, and the CCR Scientific Program Manager. \nQualifications: \nMasters or PhD preferred. Disease focus area of the laboratory include cancer (hepatocellular and renal)\, obesity\, and diabetes. Specific research areas include application of genome-wide epigenomics methods to decipher DNA methylation/hydroxymethylation defects in cancer and biochemical/cell culture-based techniques to study the regulation and interactions between chromatin remodeling factors (e.g. DNMTs\, TETs\, HMTases). Techniques commonly used in the laboratory include: quantitative RT-PCR\, western blotting\, tissue culture\, transfections\, bisulfite sequencing\, pyrosequencing\, ChIP-qPCR\, ChIP-seq\, next-gen library preparation and sequencing\, Infinium 450k arrays\, bioinformatics analysis\, and biochemical analysis of protein function. DESCRIPTION:Pfizer’s Oncology Research Unit (ORU) in La Jolla\, California\, is seeking a Scientist focused on translating basic discoveries in the field of cancer epigenetics into novel therapeutics. \nOur ideal candidate will work within a collaborative team environment to perform laboratory-based research. The candidate will employ genetic and pharmacological methods to discover and confirm candidates for novel cancer therapeutics. As a member of the group\, this person will also be involved in mechanism-of-action studies\, the interpretation and presentation of data\, and will function as an integral member of the team to advance new targets. The successful candidate will have experience in state-of-the-art techniques in epigenomic profiling (ATAC-seq\, ChIP-seq) as well as a basic understanding of epigenetic regulation in the tumor microenvironment. \n\nRole Responsibilities \nWork independently and as part of project teams to design\, execute\, analyze/interpret\, and refine experiments ranging from biochemical and molecular assays\, in vitro and in vivo target modulation in model systems\, in vitro pharmacology\, biomarker assay development\, and clinical sample analysis. \n\nDepending upon the stage of project\, specific duties to include: \n\nApplication of genetic and pharmacologic approaches to modulate drug targets in vitro\, and assess functional impact on cancer initiation and progression. DESCRIPTION:The Ntziachristos laboratory studies the mechanistic aspects of oncogenesis with an emphasis on transcriptional and epigenetic regulation of acute leukemia. We study how oncogenes interact with each other and with epigenetic modulators to influence gene expression programs as well as how their function is related to tri-dimensional (3D) structure of the nucleus and other biological aspects of cancer cells\, like metabolism. Recent studies by our group focus on how active deubiquitination controls aspects of oncogenesis and leads to drug resistance in leukemia. To address these questions we use high-throughput molecular and cell biology techniques like RNA-Seq\, ChIP-Seq\, 4C-Seq and HiC\, fluorescent in situ hybridization\, biochemical analysis e.t.c.\, in cell lines and primary cells of human origin and tissues of mouse models of disease. In addition to understanding cancer biology these finding help us design and test targeted therapies in preclinical models of leukemia. \nHighly-motivated and collaborative scientists\, that hold a Bachelor\, Master’s or Ph.D. degree\, and with a strong track record and expertize in Bioinformatics\, with a focus on RNA biology and the analysis of splicing phenomena\, computational systems biology or related fields are encouraged to apply. The candidates should have knowledge of gene expression biology and of high-throughput methodologies such as RNA-Seq\, ChIP-Seq\, HiC\, and 4C-Seq. The team utilizes high throughput bimolecular assays\, deep cellular phenotyping\, single cell genomic\, proteomic\, chromatin conformation/epigenetic and computational and bioinformatics analyses and advanced somatic cell genome modifications with CRISPR technologies in induced pluripotent cells to develop and test human cell models of human disease. Our Human Phenotyping and Translational Informatics Cores which are supported through NIH P30 and U19 mechanisms are available as well as there is access to high performance computational resources both in-house and through cloud-based resources.  \nThis is a mentored position for postdoctoral training.
