DESCRIPTION:Dr. Hari’s laboratory at Huntsman Cancer Institute and College of Pharmacy under Therapeutics Accelerator program is seeking a highly motivated Research Technician with expertise in conducting various in vitro cancer biology and drug screening assay for the kinase\, epigenetic and immune related protein targets. Interested candidate will be working on the biological evaluation of novel chemical entities (NCE) for use in cancers research and treatment. Research Technician\, you will be technically responsible for working on the PI and his labs chemist fellows to support in vitro cell-free and cancer cell-based efficacy screening assays utilizing available compounds as and when made by chemist fellows. This will entail detailed reading of the literature reported assays\, validation using reference control compounds\, associated methods\, existing assay protocols\, and alos utilizing vendor specific reagents/kits etc.\, and then planning\, execution\, optimization and analysis of the assay results. The compound effects in those assays and presenting the results in group meetings\, then supporting sections of grant proposals\, publications\, and lab supervision. \n\nResponsibilities \n\nCell-free target binding assays; kinase glo\, thermal shift and FP\, FRET\, AlphaScreen\, AlphaLisa\,\nIC50\, EC50\, Ki\, Kd\, Km\, substrate competitive\, reversibility and time dependent read out data analysis and handling expertise\nCell-based\, cancer cellular profiling and synergy experiments handling expertise are part of the job description\nAdditional experience in cancer biology investigations such as conducting standard biochemical/binding or other relevant target specific mechanistic assays such as Flow Cytometry\, NK\, B cell\, immune cell killing\, cytokine release\, Macrophage\, other immunoassays and working experience on radioligand binding assays is a plus.\nAbility to clearly communicate ideas verbally and through presentations.\n\nMinimum Qualifications \nBachelor’s degree in biology\, chemistry\, or other scientific disciplines in a related field or equivalency required; knowledge of complex laboratory techniques\, equipment\, terminology\, materials and substances. Skilled in the use of laboratory equipment\, ability to create and analyze statistical calculations and prepare reports required; and demonstrated human relations and effective communication skills also required. DESCRIPTION:The proposed position will be located at the Dermatology Research labs\, specifically in the DiNardo Lab. The dermatology labs are located at the Altman Clinical and Translational Research Institute (ACTRI) at UC San Diego\, established in 2010 as part of a national consortium of 60 medical research institutions created to energize bench-to-bedside efforts. The structure was opened in 2016 and encompasses 359\,000 square feet of offices\, state-of-the-art laboratories and clinical space (including a research pharmacy) and houses a multitude of distinct research endeavors as described before. \nThe Lab Assistant will engage in assisting postdoctoral fellow and senior scientists working on skin innate immune projects related to microbiome and immune-tolerance. Under supervision\, the lab assistant will assist with preparation and execution of experiments using biological and chemical materials. Tasks will include preparation of standardized media and reagents; upkeep of laboratory apparatus and facilities; ordering and organization of laboratory supplies; record-keeping and organization of experimental outputs on the laboratory computer notebook and databases. \nThe assistant will have an opportunity to observe and aid senior laboratory members with experiments involving cell culture\, PCR\, specimen sectioning\, molecular cloning\, genotyping and gel electrophoresis. \nMINIMUM QUALIFICATIONS\n\nGraduation from high school or a GED and one (1) year of laboratory experience or two (2) years of college including courses in the biological sciences; or an equivalent combination of education and experience.\nPossess the knowledge and abilities essential to the successful performance of the duties assigned to the position.\nProven experience doing standard and routine laboratory tasks. SUMMARY:Research Opportunities - Medical Oncology
DESCRIPTION:There is an opportunity for research in the laboratory of Dr. Jieya Shao. The central theme of the lab is unravelling novel cancer biology with the overarching goal of developing effective therapies. Two main projects are currently ongoing primarily in the context of breast cancer: \n1. “Moonlighting” functions of the actin-binding protein profilin-1 (Pfn1) in transcriptional and epigenetic regulation. As an essential eukaryotic protein\, the mechanisms behind the paradoxical anticancer effects of profilin-1 have remained mysterious for nearly two decades. Our recent studies suggest that the anticancer activities of Pfn1 may stem from its poorly understood nuclear functions which become deregulated in cancer due to an alteration of its nucleocytoplasmic trafficking. We are currently focusing on defining its novel roles in transcriptional control and epigenetic regulation in the context of both estrogen receptor positive and triple negative breast cancer by utilizing multidisciplinary approaches including biochemistry\, molecular and cellular biology\, proteomics\, mouse xenograft models\, and next generation sequencing.\n2. The role of VCP in protein homeostasis\, DNA damage response\, and chemotherapy efficacy of cancer. Valosin-containing protein (VCP) is an evolutionarily conserved and highly abundant AAA+ ATPase involved in diverse cellular processes. Functioning as a “segregase”\, VCP maintains protein homeostasis by extracting polyubiquitinated proteins from various cellular locations or complexes for turnover. Numerous studies have now linked VCP to chromatin-associated protein degradation and genome stability maintenance particularly in the context of DNA damage response. Nevertheless\, detailed understanding of its mechanism of action\, its mode of (de)regulation in cancer\, and its relevance to chemotherapy response is lacking\, and we are currently investigating all these questions. \n\n\nPreferred Qualifications\n\n\n\n\n\n\n\n\nCandidates with general understanding of biochemistry\, molecular and cellular biology and prior experience in mammalian cell culture\, Western blotting\, DNA/RNA isolation\, mouse handling and surgery are preferred.\nKnowledge in genetics/epigenetics and experience in quantitative PCR and chromatin immunoprecipitation (ChIP) is a plus\, so is the skill in data mining and computational analysis.\nMust have troubleshooting skills\, independent analytical thinking and research planning skills.\nDemonstrated ability to work independently and communicate with the laboratory staff and investigators.\nAbility to maintain complete and organized records/reports.\nWorking knowledge of computer software and general lab equipment.

SUMMARY:Dynamics of Epigenetic Regulation: Mechanisms and Beyond
DESCRIPTION:The Gordon Research Seminar on Epigenetics is a unique forum for graduate students\, postdocs\, and other junior scientists to present and exchange new data and cutting-edge ideas. The focus of this meeting is on recent advances in our understanding of the dynamic nature of epigenetic regulatory mechanisms across phyla. In particular\, we will focus on how epigenetic alterations can influence phenotypic plasticity\, development\, reprogramming\, and transgenerational inheritance. The GRS will include a keynote address by Dr. Bradley Cairns\, as well as a career panel discussion with panelists from academic and non-academic fields. \nApplication Instructions\nThe seminar will feature approximately 10 talks and 2 poster sessions. All attendees are expected to actively participate in the GRS\, either by giving an oral presentation or presenting a poster. Therefore\, all applications must include an abstract. \nThe seminar chair will select speakers from abstracts submitted by April 20\, 2019. Those applicants who are not chosen for talks and those who apply after the deadline to be considered for an oral presentation will be expected to present a poster. In order to participate\, you must submit an application by the date indicated in the Application Information section above. \nProgram Format\nGordon Research Seminars are 2-day meetings which take place on the Saturday and Sunday just prior to the start of the associated GRC. The GRS opens with a 1-hour introductory session on Saturday afternoon\, followed by a poster session\, dinner and a 2-hour session in the evening. Sunday morning begins with breakfast and is followed by another 2-hour session\, a second poster session\, and lunch. A final 1-hour session takes place just after lunch\, and the associated GRC begins later that evening. \nAn outline of the program components for this GRS is displayed below. The seminar chair is currently developing their detailed program schedule\, which will include the speakers they select from submitted abstracts\, in addition to any additional components outlined below.
