SUMMARY:Research Opportunities - Medical Oncology
DESCRIPTION:There is an opportunity for research in the laboratory of Dr. Jieya Shao. The central theme of the lab is unravelling novel cancer biology with the overarching goal of developing effective therapies. Two main projects are currently ongoing primarily in the context of breast cancer: \n1. “Moonlighting” functions of the actin-binding protein profilin-1 (Pfn1) in transcriptional and epigenetic regulation. As an essential eukaryotic protein\, the mechanisms behind the paradoxical anticancer effects of profilin-1 have remained mysterious for nearly two decades. Our recent studies suggest that the anticancer activities of Pfn1 may stem from its poorly understood nuclear functions which become deregulated in cancer due to an alteration of its nucleocytoplasmic trafficking. We are currently focusing on defining its novel roles in transcriptional control and epigenetic regulation in the context of both estrogen receptor positive and triple negative breast cancer by utilizing multidisciplinary approaches including biochemistry\, molecular and cellular biology\, proteomics\, mouse xenograft models\, and next generation sequencing.\n2. The role of VCP in protein homeostasis\, DNA damage response\, and chemotherapy efficacy of cancer. Valosin-containing protein (VCP) is an evolutionarily conserved and highly abundant AAA+ ATPase involved in diverse cellular processes. Functioning as a “segregase”\, VCP maintains protein homeostasis by extracting polyubiquitinated proteins from various cellular locations or complexes for turnover. Numerous studies have now linked VCP to chromatin-associated protein degradation and genome stability maintenance particularly in the context of DNA damage response. Nevertheless\, detailed understanding of its mechanism of action\, its mode of (de)regulation in cancer\, and its relevance to chemotherapy response is lacking\, and we are currently investigating all these questions. \n\n\nPreferred Qualifications\n\n\n\n\n\n\n\n\nCandidates with general understanding of biochemistry\, molecular and cellular biology and prior experience in mammalian cell culture\, Western blotting\, DNA/RNA isolation\, mouse handling and surgery are preferred.\nKnowledge in genetics/epigenetics and experience in quantitative PCR and chromatin immunoprecipitation (ChIP) is a plus\, so is the skill in data mining and computational analysis.\nMust have troubleshooting skills\, independent analytical thinking and research planning skills.\nDemonstrated ability to work independently and communicate with the laboratory staff and investigators.\nAbility to maintain complete and organized records/reports.\nWorking knowledge of computer software and general lab equipment.

DESCRIPTION:In the Kriwacki lab we are passionate about understanding how phase separation drives the condensation of biomolecules to create liquid-like bodies that mediate diverse biological processes. Importantly\, aberrant biomolecular condensation is associated with several devastating human diseases\, such as cancer and neurodegeneration.  A new initiative in the laboratory addresses the role of aberrant phase separation as a driver in cancer and we seek an analytical\, critical thinking\, self-driven cell biologist to join our team. DESCRIPTION:The Beltran laboratory at Dana-Farber Cancer Institute and Harvard Medical School is focused on the study of treatment resistance in prostate cancer. We utilize a variety of molecular biology and genomic and epigenomic approaches to understand prostate cancer disease progression\, therapy resistance\, and tumor heterogeneity both in patients and in preclinical models. We are particularly focused on the emerging subset of advanced prostate cancers that lose androgen receptor dependence as a mechanism of treatment resistance\, associated with lineage plasticity and a change in clinical and pathologic characteristics towards a small cell/neuroendocrine phenotype. Current projects include: 1) Defining and validating the essential genes during neuroendocrine prostate cancer progression using genome-scale CRISPR-Cas9 screening approaches to identify novel therapeutic targets; 2) Understanding the genetic\, epigenetic\, and phenotypic changes that drive cancer progression through bulk and single cell molecular analyses of tumor biopsies and patient-derived models; 3) Elucidating the mechanism\, timing\, and cooperation of identified drivers including MYCN\, AURKA\, EZH2\, RB1\, and others; 4) Characterization and preclinical testing of novel epigenetic and immune -based therapeutic approaches and combination strategies to treat\, revert\, or prevent non-AR driven resistance. Research in the laboratory ranges from basic mechanistic investigation of cancer gene function to highly translational genomic and functional studies of patient-derived tissues from clinical trials. Team members work in a highly collaborative environment that includes laboratory investigators\, clinical investigators\, and computational biologists. We are seeking an enthusiastic\, dedicated and hardworking post-doctoral scientist to join our team. The successful candidate will have a strong interest in cancer biology and therapeutics with experience in molecular biology\, biochemistry\, genetics/genomics or biochemistry. DESCRIPTION:The Hackensack Meridian Health Center for Discovery and Innovation is offering a position for postdoctoral studies in the field of cancer signaling and gene expression in the laboratory of Dr. Byungwoo Ryu.  The goal of the laboratory’s research program is to unravel the cancer epigenome regulating mechanisms which can be targeted for the sensitization to the precision therapies in the hope of translating these therapeutic modalities to have long-term clinical benefit. The Ryu laboratory focuses on investigating chromatin remodeling enzymes\, such as histone and DNA modifiers and gene transcription factors as potential targets for overcoming the resistance of oncogene-targeting therapies and enhancing the objective therapy response rate of immune check-point blockade therapies.  The Ryu laboratory uses chemical genetics approaches including small molecule inhibitors\, genome/epigenome-wide profiling\, gene-editing technology\, and animal models. Successful postdoctoral candidate will have a strong interest in cancer biology\, epigenetics\, and immuno-oncology.  The candidate will perform in vivo animal models and in vitro analyses. The postdoctoral fellow is expected to understand the project and work independently taking responsibilities in the development and execution of experimental procedures to successfully advance the research project. The successful candidate will be responsible for preparing data for presentations\, progress reports\, and the preparation of manuscripts and posters. DESCRIPTION:Rutgers\, The State University of New Jersey is seeking a Post Doc Fellow for the Cancer Pharmacology Department at Rutgers Cancer Institute of New Jersey. \nThe primary purpose of the Postdoctoral Appointee position is to conduct research investigation into the characterization of oncogenic and tumor suppressor enhancer regions\, as well as on the dissection of the interplay between metabolism and epigenetics in T-cell Acute Lymphoblastic Leukemia (T-ALL) using mouse models under the supervision of the Principal Investigator. The Post Doc Fellow will perform research\, experiments\, and laboratory techniques of a specialized nature under direct supervision of the Principal Investigator. \nAmong the key duties of this position are the following:\n• Conducts research experiments to understand the role and mechanisms of SOD1 in oxidative stress resistance and cancer cell survival as well as the interplay between metabolism and redox regulation to unveil new therapeutic strategies. \n• Prepares written reports of the research experiments. \n• Prepares grant proposals based on the research conducted. \n• Gives oral presentations of the research. \n• Performs other related duties as assigned.
